  • Protein - small molecule interactions
Protein aggregation studies: Aggregation studies currently underway involve human serum albumin, lysozyme and the amyloid peptide fragment Ab25-35. Effects of a static electric field on the disruption of fibrils have been reported. Work with polyphenols and surfactants and their effects on the fibrillation process of proteins are in progress. The disruption of the fibrillation of amyloid fibrils, the hallmark of neurodegenerative diseases in the presence of external additives is currently underway.

Studies with eye lens proteins: Modified γ-crystallin (HGCc) from the discarded cataractous emulsion of patients has been isolated and characterized for modifications during cataract formation. Studies showed that the major polyphenol present in green tea, EGCG was able to prevent tryptophan oxidation of the cataractous ocular lens human γ-crystallin in presence of H2O2. The discarded mass is being used to prepare protein nanoparticles and films for applications in drug delivery, wound healing etc. A patent has been awarded for the use of this novel protein source in the preparation of the nanoparticles.

Protein ligand binding: Studies with ribonucleases such as angiogenin, a potent inducer of blood vessel formation, indicated that the process of angiogenesis, the capacity to form blood vessels, was inhibited by polyphenols present in green tea. The polyphenols were also shown to behave as noncompetitive inhibitors of Ribonuclease A. Other inhibitors of the proteins are being developed in collaboration. The effects of oxidative stress on the proteins and methods to combat them are in progress.
